SOP for Media Preparation

1.0 OBJECTIVE :
  • To provide a Procedure for Preparation and Destruction of Media for Microbiological analysis.
2.0 SCOPE :
  • This procedure applies only for preparation and destruction of media used in Microbiological analysis.

5.0 PROCEDURE :

5.1 PREPARATION OF MEDIA:

  • Prefer readymade media packs of Hi-Media for use.
  • Use clean conical flask for preparation of media.
  • Use distilled water/purified water for preparation of media.
  • Read the instructions given on the media pack, prepare the media accordingly.
  • Calculate the quantity of media to be weighed and weigh the quantity, record the weight and transfer to flasks, add small quantity of distilled water, shake the flask till it gets  completely dissolved and add total quantity of distilled water.  Mix well the content of flask.
  • Check the pH of it before sterilization & record the observed pH.  The acceptance Criteria of pH is ± 0.2 of value available on pack. 
  • In case of deviation investigate and take appropriate action.
  • Wrap the mouth of conical flask with aluminum foil.
  • Keep these flasks in the autoclave.
  • Keep a sterilization strip along with the preparation load and start the autoclave at 121°C for 15 min, 15 lbs.
  • Allow the pressure to come down to zero then open the lid and remove the sterile preparation load.
  • Prepare XLDA media as given on the media pack label and do not autoclave.
  • Check the pH of media after autoclaving.
  • Record the observed pH.
  • Excess overheating can loss the properties of media ingredients, follow exact time, temperature & pressure during autoclaving.
  • Keep positive and negative controls for the batch of media to check proper growth promotion of the respective organisms and negative control to check complete sterility of media during analysis.
  • Maintain the media preparation record.
