SOP for Colony Counter

1.0 OBJECTIVE :
  • To provide a procedure for Cleaning and Operation of Colony Counter.
2.0 SCOPE :
  • This procedure is applicable to count the number of colonies in Microbiology Section using DBK make Instruments.
3.0 RESPONSIBILITY :
  • Lab assistant under supervision of microbiologist & microbiologist
4.0 ACCOUNTABILITY:
  • Head of Department
5.0 PROCEDURE :
5.1 Cleaning :
  • De-dust the external surface of colony counter and magnifier with lint free duster.
  • Clean the external surface of colony counter with 70 % v/v Iso Propyl Alcohol.
  • Frequency: Daily
5.2 Operation:
  • Connect the colony counter to the main supply and switch “ON” the power switch.
  • Switch “ON” the mains switch of the equipment.
  • Digital display show “0000”, if the digital display is not showing “0000” then press the reset button, digital display show “0000”.
  • Keep the petri dish on illuminating surface.
  • Adjust the magnifying glass to view clear colonies present on the petri dish.
  • Press the tip of the pen on the colonies present on the petri dish and count the number of colonies.
  • Record the number of colonies.
  • Remove the plate from the wolffhuegel grid plate.
  • Press the reset button.
  • Switch off the mains after completion of work.
