SOP for Microscope
1.0 OBJECTIVE :
- To provide a procedure for Cleaning and Operation of light Microscope.
2.0 SCOPE :
- This procedure is applicable to Microscope (Standard Model Labomed Vision 2000).
3.0 RESPONSIBILITY :
- Lab assistant under supervision of microbiologist & microbiologist
4.0 ACCOUNTABILITY:
- Quality Control Head
5.0 PROCEDURE :
5.1 Cleaning :
- Disconnect the instrument from electrical connection.
- Clean the instrument with soft cloth.
- Clean the eyepiece, objective and optical parts (mirror, lens) using soft cloth or tissue paper.
- Also clean with Cotton dipped in acetone and dry in air.
5.2 Operation:
- Switch on the main switch of microscope.
- Adjust the high power objective, by rotating the revolving nose piece in position.
- Position the slide to bring into the exact center of the light source.
- Focus high magnification objective lens on the center of the sample.
- Turn the coarse adjustment knob to adjust the stage.
- Bring the object into focus by adjusting with the coarse Knob. Focus the sample for better resolution by using fine adjustment knob.
- Adjust the diaphragm level to decrease or increase the light intensity to produce the clear and sharp image.
- After completion of observation, remove the slide and clean the objective lens with cotton soaked in xylene.
- Switch off the main switch of microscope.
